The former Cardiff City player and Wales international will join the Owls on July 1 when his contract with the Bluebirds expires.
His arrival follows the sacks of central defender Ben Heneghan and experienced goalkeeper David Stockdale.
Vaulks began his career with Tranmere Rovers before moving north of the border to Falkirk.
He returned south to the Millers in 2016 and rose to fame during his time at the South Yorkshire club, playing 125 games and winning Player of the Year in his second season as his side won promotion to the Championship in 2017-18. .
Following the Millers’ relegation at the end of the following season, Vaulks moved to Cardiff for a seven-figure fee and made over 100 appearances in his three seasons at the club.
Wednesday are also on the hunt for two of Vaulks’ former Millers colleagues, Michael Smith and Michael Ihiekwe, whose deals expire at the end of this month, with the pair yet to agree new deals.
The Millers are still struggling to keep both players and convince them to stay at AESSEAL New York Stadium, despite offers from other venues, especially on Wednesday.
Confident that the Championship newcomers will have completed the additions of two full-backs and potentially additions in the attacking and centre-half departments by the end of the week, Millers manager Paul Warne told the BBC: “With Regarding Smudge (Smith) and Iky (Ihiekwe), I know that they are well regarded in our football club, we have done everything possible with the support of the president to try to keep them.
