The A1 southbound is closed between exits 47 and 48 near Boroughbridge.
The accident occurred at 7.48 am this morning (June 12) and involved only one vehicle.
North Yorkshire Police said: “The driver and passengers of the vehicle are believed to have serious but not fatal injuries.”
Anyone traveling in the area is urged to avoid the A1 due to heavy congestion.
The police also asked anyone who witnessed the accident to contact the police at 101, passing information to the force control room.
